Abstract

Administration of Corynebacterium parvum in combination with localized irradiation has been shown to be more efficient than radiation treatment alone in prolonging the survival time of methylethrane induced tumor in C3H mice. Fractionated low dose total body irradiation has been demonstrated to be a promising therapy for patients with chronic lymphocytic leukemia. The purpose of this study was to determine the effect of the combined treatment of total body irradiation and C. parvum on Friend virus induced leukemia in C57BL/6 mice. 125I-labeled FBL-3 cells were injected intraperitoneally into mice. Intraperitoneal injection of C. parvum and and single dose total body irradiation were given to the mice immediately after tumor inoculation. Fractionated total body irradiations were given to the mice every day .for 10 days after tumor inoculation. For two injections of C. parvum the first injection was given 16 days before, the second one on the day of tumor inoculation. The fate of the labeled tumor cells was measured by counting the 125I-retention of individual live mice in a well-type crystal scintillation counter. The percent of 125I-activity remaining in the mouse was taken as an indicator of the effectiveness of the treatments. The results indicated that the combined treatment of a single dose total body irradiation and intraperitoneal administration of C. parvum enhanced the the elimination of the tumor more efficiently than the separate treatments. The combined treatment of fractionated low dose total body irradiation and intraperitoneal administration of C. parvum also enhanced the elimination of the tumor more efficiently than the fractionated radiation treatment alone; but the effect of the combined treatment was only slightly greater than the C. parvum treatment alone. Two injections of C. parvum in combination with fractionated low dose irradiation greatly enhanced the elimination of the tumor over no treatment at all, but only slightly over the treatment of two injections of C. parvum alone.
