Abstract
The purpose of this study was to identify the early hard tissue formed in experimental pulp exposures capped with calcium hydroxide, mineral trioxide aggregate (MTA) or bone morphogenetic protein-7 (BMP-7) using dentin sialoprotein (DSP) as a marker. The pulps of eighty-five maxillary first, second and third molar teeth from twenty male rats were experimentally exposed. The exposed pulps were capped with calcium hydroxide followed by restoration with amalgam (n = 20), with MTA alone as a pulp capping agent and final restoration (n = 20) or with BMP-7 followed by restoration with MTA (n = 20). Ten teeth with Class 1 occlusal preparations, no exposure and no restoration served as positive controls. Ten teeth that received pulp exposures and no restoration served as negative controls. Five untreated third molars served as additional controls. The animals were sacrificed at two and eight weeks. The specimens were prepared and evaluated histologically and with immunohistochemistry utilizing polycolonal antibodies raised against rat dentin sialoprotein (DSP). Pulps capped with calcium hydroxide demonstrated irregular hard tissue formation that did not stain for DSP. Pulps capped with MTA formed hard tissue that demonstrated significantly more immunostaining for DSP when compared to calcium hydroxide and BMP-7 (p = 0.0001 and p = 0.0031 respectfully). MTA capped pulps also demonstrated significantly more complete bridging when compared to calcium hydroxide and BMP-7 (p = 0.0008 and p = 0.008 respectfully). Pulps capped with BMP-7 demonstrated a hard tissue that was bone-like in appearance and devoid of DSP staining.
